Callus culture for biomass production of milk thistle as a potential source of milk clotting peptidases Electron. J. Biotechnol.

The objective of this work was the optimization of the conditions of in vitro culture for callus production of Silybum marianum (L.) Gaertn. (Asteraceae). Sections of cotyledons, previously disinfected by washing successively with ethanol 70º, NaClO (10% w/v) and Tween 20 (0.05% v/v) and rinsing with sterile distilled water, were used as explants. For its initial culture, B5 medium supplemented with BA and 2,4-D solidified with phytagel was used, and a 63% survival was achieved. To obtain callus, two solid media were assayed (S1 and S2) using B5 medium supplemented with growth regulators (BA and 2,4-D or NAA and BA, respectively). Callus induction and plant regeneration of the endemic Astragalus nezaketae in Turkey Electron. J. Biotechnol.

A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. The combinations and concentrations of PGRs were shown significant variations for the frequency of callus formation, appearence of callus and the potential of callus differentiation. Comparative Microbicidal Efficacy of Fractionated Extracts from In Vitro and In Vivo Raised Cells of Tinosporacordifolia Against MDR Pathogens BABT

The present study was conducted to explore the hidden potential of natural products synthesized in the medicinal plant Tinosporacordifolia. This plantis prioritized by National Medicinal Plant Board, New Delhi. Leaf and inter nodal segments were inoculated on MS Medium fortified with IBA (1.0 mg/L) produced callus after four weeks. The calli were brown due to phenolic substance secreted by the explant. This problem was overcome by using adjuvant PVP (0.1%). ABSTRACTThe present investigation was aimed to detect the specific polypeptide(s) appeared during the sequential stages of differentiation. Among different explants, only nodal explants showed good results for callusing. Depending on the fresh and dry weight, best callus growth was observed on MS medium supplemented with NAA (2.5 mg/L) inDioscorea alata and 2, 4-D (2.0 mg/L) inD. deltoidea, respectively. To define preliminary aspects of walnut (Juglans regia L.) callogenesis in indirect propagation systems, this work analyzed callogenic induction in adult leaves of walnut in broadleaf trees (BTM), Murashige and Skoog (MS) Driver Kuniyuki Walnut (DKW) and Woody Plant (WPM) media. Leaf segments of 1 cm², previously sterilized, were placed in each culture medium, supplemented with 21.4 µM naphtalene acetic acid (NAA) and 8.8 µM 6-benzylaminopurine (BAP). Campomanesia adamantium is a native plant species of Brazilian Cerrado with diverse economic potential and great medicinal importance. Its sexual propagation is impaired by the recalcitrance of its seeds, which prevents effective and profitable propagation. Estudou-se os efeitos de auxilias exógenas e ácido bórico, no enraizamento de estacas de lichia (Litchi chinensis Sonn.). As estacas foram uniformizadas, com 25 cm de comprimento e 4 folhas cortadas pela metade. Cerca de 2,5 cm da base das mesmas foi mergulhado nos tratamentos: H2O; Boro 150 µg/ml; IBA 5.000 ppm, IBA 2.000 ppm; IBA 5.000 ppm + Boro 150 µg/ml; IBA 2.000 ppm + Boro 150 µg/ml; NAA 3.000 ppm; NAA 1.500 ppm; NAA 3.000 ppm + Boro 150 µg/ml; NAA 1.500 ppm + Boro 150 µ/g/ml. Improved method for isolation of coupled mitochondria of Araucaria angustifolia (Bert.) O. Kuntze BABT

A method for the isolation of coupled mitochondria from the callus of Araucaria angustifolia is described for the first time. Mitochondria were isolated from embryogenic callus of A. angustifolia. They were metabolically active, able to sustain oxidative phosphorylation as shown by respiratory control ratio values, which were about 2.4 when respiring on succinate as substrate. Oxygen uptake experiments, using freeze-thawed disrupted mitochondria, showed the presence of alternative rotenone-insensitive NAD(P)H dehydrogenases, which were stimulated by Ca2+. The procedure now described for the isolation of A. angustifolia mitochondria is an important new tool, allowing the investigation of mitochondrial bioenergetics and metabolism and physiology of plants.

In vitro establishment and multiplication of the Normania triphylla (Lowe) Lowe BABT

Normania triphylla is an endemic species from Madeira island (Portugal) extinct in the wild since 1991. The aim of this work was to culture the meristems of this species in vitro and to multiply its shoots in order to preserve this endangered species. The best results in terms of multiplication were obtained in Murashige and Skoog medium supplemented with 10 µM 6‑benzylaminopurine (BAP). The number of shoots, the number of nodes and the number of leaves were the most important in this medium. In vitro morphogenic response from zygotic embryos of Genipa americana Ciência Rural

ABSTRACT: The aim of this study was to evaluate the in vitro morphogenic potential of genipap (Genipa americana L.) zygotic embryos. Seeds obtained from ripe fruits had their zygotic embryos excised and inoculated in MS medium with 4.44µM of 6-benzylaminopurine (BAP) and supplemented with 0.0; 1.07; 2.14 and 3.21µM of naphthalene acetic acid (NAA). The potential of explants regeneration and the shoot length and number of leaves in plantlets were evaluated. The in vitro regeneration of genipap is possible from the conversion of zygotic embryos in a MS medium with 4.44µM BAP supplemented with 3.21µM NAA.

BACKGROUND: The aim of the present work was to examine the role of UV-C irradiation on the production of secondary metabolites (total phenolic, total flavanols, total flavonols, catechin, ferulic acid and trans-resveratrol in phenolic compounds and α-, β-, γ- δ-tocopherols) in callus cultures. Curcuma zedoaria Roscoe (zedoary) is a medicinal properties-bearing Zingiberaceae from which rhizomes are commercially exploited. The objective of this work was to establish an in vitro protocol for micropropagation and callogenesis of Curcuma zedoaria Roscoe as alternative to improve plant production, turning economically feasible the exploitation of its secondary metabolites which present medicinal properties. Physiological Responses of Callus from Gerbera jamesonii Bolus ex. Hook f. to Gamma Irradiation BABT

In the present study, in vitro mutagenesis techniques were applied to investigate the effects of gamma irradiation at 0, 10, 20, 30, 40, 50 and 60 Gy on physiological changes in callus of Gerbera jamesonii Bolus ex. Hook f. Biochemical changes in chlorophyll and soluble protein content of pre- and post- irradiated Gerbera callus were studied. Non-irradiated callus demonstrated the highest amount of chlorophyll content as compared to callus irradiated at 10, 20, 30, 40, 50 and 60 Gy. Plant regeneration after long term callus culture in clones of Asparagus officinalis L. Biocell

Callus growth and plant regeneration from long-term callus cultures were studied in two elite clones of Asparagus officinalis cv. Argenteuil, to establish a suitable protocol for a prospective in vitro selection program. Callus initiation and growth was evaluated on MS medium with 3% sucrose, 0.9% agar, 1 mg.l-1 kinetin, and three levels of 2,4-D. The highest callus relative growth was obtained on medium with 1.5 mg.l-1 2,4-D and 1 mg.l-1 kinetin.
